Dear Users, The scratch2 drive is getting full, so we need to delete older files from it. We start deleting files older than the 01st August 2024 today. NB This will include data files (eg exel data etc). Please make sure you have everything backed up, as mentioned during the training session, make multiple backups of images on the day of the imaging session. Please remember that the Scratch2 drive is for transferring FILM data only. It is not for long term storage or for users to store other data and programs. If you are working offsite then Scratch2 can be accessed either through VPN or using the remote gateway to logon to you work PC. [FILM-Users 01173] Update on Staffing and Operations for FILM Users

Dear Colleagues, I hope this message finds you well. I would like to inform you of a current substantial shortage in our staffing. As of today, I am the only staff member fully available across three sites (ICTEM, SAF, Flowers). We anticipate that within the near future, staffing levels will increase to 2-3 members. Please note that David will be unavailable for work until September 19, 2024. I apologize for any delays and inconvenience this may cause and appreciate your understanding during this period. As FILM manager, I am committed to allocating appropriate staff resources to maintain the best possible performance of our imaging services. [FILM-Users 01172] Update on FILM Facility Charging Rates

Dear Colleagues, As you know, FILM, our imaging facility, has been operating under significant economic and technological pressures, reflecting the broader situation in the UK economy. The annual inflation rates over the last four years have been as follows: - 2021: 2.6% - 2022: 9.1% - 2023: 7.3% - 2024: 3.1% (up to July) The cumulative inflation over these four years is approximately 24.2% (23.08%)<https://www.bankofengland.co.uk/monetary-policy/inflation/inflation-calcula…>: [cid:3bffd417-5ac5-42fc-9c45-0e25fd70814a] These inflationary pressures have led to increased costs in staff expenses, service contracts, consumables, and necessary investments in IT infrastructure within the SAF and ICTEM Buildings. Despite these challenges, our charging rates<https://www.imperial.ac.uk/medicine/facility-for-imaging-by-light-microscop…> have remained fixed over the last four years. We have tried to compensate by increasing our income through the acquisition of new equipment and intensifying our workload, which has resulted in income growth each year, including a 10.9% increase in 2023. However, due to limitations in staff numbers and other factors, it is no longer feasible to maintain the current charging rates. In the near future, we will be developing a new charging schema to address these challenges. Thank you for your understanding and continued support. Dear Colleagues, We are pleased to announce that the Leica UK Team is organizing the next workshop in our Stellaris<https://www.leica-microsystems.com/products/confocal-microscopes/p/stellari…> applications series on Tuesday, 3rd September. The topic for this session will be “Autonomous Microscopy.” Autonomous Microscopy enables highly economical operations for daily laboratory work, allowing advanced experiments that would otherwise require significant manual effort or be impossible without automated procedures. This approach includes a rare event detection workflow powered by AI-based microscopy, which facilitates the automated detection of rare events without human intervention and the complete automation of complex microscopic workflows. In this autonomous workflow, low-resolution 2D overview images are initially generated and immediately transferred to the connected AI-based image processing system (Aivia<https://www.leica-microsystems.com/products/microscope-software/p/aivia/>). This system detects rare events, previously defined by the operator, using a pixel classifier and sends the event coordinates back to the imaging system for high-resolution and 3D data stack scanning according to the operator's specifications. The result is highly specific data on the object of interest, generated in statistically relevant quantities due to the automation process. Please find the program of the event attached. [FILM-Users 01170] Introducing the Cell DIVE imager

Dear All, We are thrilled to announce that the Cell DIVE imager<https://www.leica-microsystems.com/products/light-microscopes/p/cell-dive/> is now available for training sessions. The system is an automated microscope designed specifically for multiplexed experiments. It allows multiplexed tissue analysis for investigation of the expression of multiple biomarkers and the cellular and spatial localization of biomarker expression within a tissue. Please find attached the resources available in the Cell DIVE room (ICTEM room 304B). In addition to the imager and robot, the room is equipped with an antigen retrieval module, a sample preparation bench with reagents (excluding antibodies), a barcode printer, and an analysis PC. To give you an idea of what the Cell DIVE workflow looks like, you start by doing a scan with a 2x and a 10x objective to define the regions of interest, and then proceed to image the biomarker stainings with a 20x objective. You can use 4 antibodies (+DAPI) per round. Between each round you need to inactivate the dyes and image autofluorescence. We have indicative times of acquisition per round using the 20x imaging: · 1 cm x 1cm region (225 FOV): 12 minutes · 4.5 cm x 2 cm region (2584 FOV): 132 min (2h12) Leica recommends using validated antibodies from CST and Abcam. Based on the available resources, we have decided to remove the weekly limits on microscope usage while maintaining daily limits. This change will help users with long-term projects maximize their data collection without delays. Dear users, >From next week I will be working from home two days a week, usually on Mondays and Wednesdays, to concentrate on administrative tasks. I will be able to offer support remotely by email. I will schedule trainings for two other days, while the last day I will be concentrating on IT/software issues that need solving. I will let you know once this is done. You might also have noticed that rooms 408, 409 and 410 (as well as room 517 where the Elyra is) are now booked for cleaning every Tuesday from 6am-9am. If you have an overnight imaging session running at this time, you will have to stick a sign to the door so the cleaners do no enter and disturb your experiment.
