Dear FILM Users and Colleagues, We sincerely thank you for your loyalty, use of the facility, and continued support. At FILM, we strive to meet your needs and offer the best possible service. The FILM team, under Andreas' and my leadership, has been working under pressure, especially in the area of user support. Below are some key statistics on staff and user numbers since 2020: Year Effective Staff Users Groups % Over Norm 2023 3 429 156 286% 2022 3.4 426 168 250% 2021 3 383 149 255% 2020 3 321 129 214% >From December 2024, we anticipate temporarily having only two staff members—David and myself—working at the facility. However, we are making every effort to attract new talents and hope to have additional staff soon. To enhance our support, I am considering organizing a microscopy and imaging course. I have previously taught similar courses in Glasgow and Oxford, with a focus on postdocs, PhD students, and junior imaging facility managers. The course spanned 16 hours, and I also edited the textbook Principles of Light Microscopy: From Basic to Advanced (2022)<https://link.springer.com/book/10.1007/978-3-031-04477-9>, which has been downloaded 28K times. [FILM-Users 01179] ADVISORY: ICTEM New 86X objective automated water dispenser on HCF3

Hi all, This is an email to advise you that we have recently had our 86X water objective returned after it was repaired, it has been reinstalled on HCF3 with the addition of an automated water dispenser - this is to facilitate high throughput plate imaging. [cid:cb25f4a7-134d-4948-84ce-a0d18e979c18] Some users may remember interference with the cable attached to the 86X objective and the sample holder - it has been fixed into a position to minimise interference, but we urge you to handle it with care and get in touch if you have any issues. In addition to the cable, is the new tubing from the objective to the water reservoir, located at the far right of the microscope body. Please do not remove the tubing and report any instances of detachment/severing to FILM staff - this has already occurred once. On several instances the reservoir has become detached from its magnetic base with the movement of the objective turret - we ask that if this happens you place it back at the end of your session, in order to have smooth usage of this new feature we ask that all users report any issues and be respectable of facility equipment. The 86X objective takes up 2 spaces in the turret, as a result the 63X water which was in place has been relocated back to its place on HCF2 - we apologise for any inconvenience caused. We are awaiting a demo by the Leica applications team to go over the operating instructions of the dispenser in LASX and will update you all once we have compiled a guide. In the meantime Leica engineers have given us the go ahead that the objective can be used by manual addition of water for the time being. If you are interested in doing high throughput automated (glass bottomed) plate imaging for either confocal or STED imaging, please get in contact with us so we can keep you informed. Dear users, Prof Dylan Owen from the University of Birmingham is giving a talk at the ICTEM building on “Mapping the nanoscale organisation of proteins in cells” Professor Owen's lab works in three main areas: one, the development of methods for superresolution fluorescence microscopy, with particular focus on statistical image analysis techniques. Here, they have developed a number of avenues for analysing protein distributions (nanoscale clustering) from single-molecule localisation super-resolution microscopy data. Two, studying the biophysics of cell membranes, in particular protein and lipid spatio-temporal organisation at the plasma membrane. They are particularly interested in the mechanisms that regulate membrane protein organisation including protein-protein interactions, the cortical actin cytoskeleton, lipid domains and membrane topography and three, understanding the role of nanoscale organisation in regulating signalling pathways, especially at the T cell immunological synapse. T cells must perform a delicate balance – they should activate when they detect minute amounts of foreign, potentially pathogenic peptides, but not activate in response to the body’s own proteins. Much of the proteins responsible for making such decisions show complex nanoscale organisation, and aberrations in that architecture has links to autoimmune diseases including arthritis, lupus and diabetes. Dear users, Nature Conferences are organising a free one-day event about Spatial Biology on 29th October ------------------ Methods for analyzing the biological complexities of tissues with single-cell and high spatial resolution have revolutionized the study of healthy tissues as well as our understanding of complex diseases like cancer. Among these so-called ‘omics strategies, spatially resolved transcriptomics and highly multiplexed tissue imaging have emerged as especially informative. By revealing the molecular properties of cells and their spatial arrangement and distribution, these approaches allow researchers to investigate complexity and organization at the cellular, tissue and organ scales, and to build reference atlases of individual tissues.
