[FILM-Users 01170] Introducing the Cell DIVE imager
Dear All, We are thrilled to announce that the Cell DIVE imager<https://www.leica-microsystems.com/products/light-microscopes/p/cell-dive/> is now available for training sessions. The system is an automated microscope designed specifically for multiplexed experiments. It allows multiplexed tissue analysis for investigation of the expression of multiple biomarkers and the cellular and spatial localization of biomarker expression within a tissue. Please find attached the resources available in the Cell DIVE room (ICTEM room 304B). In addition to the imager and robot, the room is equipped with an antigen retrieval module, a sample preparation bench with reagents (excluding antibodies), a barcode printer, and an analysis PC. To give you an idea of what the Cell DIVE workflow looks like, you start by doing a scan with a 2x and a 10x objective to define the regions of interest, and then proceed to image the biomarker stainings with a 20x objective. You can use 4 antibodies (+DAPI) per round. Between each round you need to inactivate the dyes and image autofluorescence. We have indicative times of acquisition per round using the 20x imaging: · 1 cm x 1cm region (225 FOV): 12 minutes · 4.5 cm x 2 cm region (2584 FOV): 132 min (2h12) Leica recommends using validated antibodies from CST and Abcam.
