Dear microscopists, we are approaching the end of the academic year, and so is this year's microscopy day, again a whole day packed with information about everything related to microscopy. Those who have attended previous years will be able to tell you that it's the best opportunity to get answers to all your questions about microscopy. And even if you have already previously, we have updated our lectures and added new topics. This year, we will also finish the day with a plenary session by an invited speaker (to be confirmed) and some drinks afterwards. After a successful start, we will also have the Microscopy Match Maker & poster session again - an excellent opportunity to form collaborations, get ideas and feedback, or just see what other people are doing. The presentations are only 3 minutes / max. 3 slides, so not much to prepare, so please make sure you put your name down (emailing to <film-service(a)imperial.ac.uk>). And PIs, please encourage your students and postdocs to participate, to show their / your research and connect with other labs. [FILM-Users 00357] FILM club Friday 31 May 1pm: Extracting complex information from images using Definiens Developer XD

Dear microscopists, in next week's FILM Club I will give an overview about the application of Definiens Developer XD for the analysis of complex microscopy data in the facility. Definiens Developer XD is a powerful software tool to extract complex information from unstructured image data, way beyond the usual thresholding and filtering approaches biologists like to use. It gives biologists easy access to a large range of advanced image analysis tools, which allows to analyse images in a similar way to visual inspection, handling challenging problems like low contrast, label-free brightfield images, high noise and background, object tracking, etc. Tools like edge detection or various noise filters help dealing with limitations of image acquisition, while the problems of false positives / negatives through strict thresholds can be overcome through fuzzy logics. Once objects are identified, a few clicks produce a wealth of results like compactness, elliptic fit, border index, position, orientation, relative distances and much more. In this presentation, I will give a short overview how these tools are used for the analysis of real data produced in the facility, where we have processed up to 30,000 images in one experiment. And please note that in September we are going to organise a Definiens Developer XD training course (and an Advanced course a bit later, probably in November), so if you have challenging microscopy data you need to analyse, this FILM Club should give you an idea whether this is your solution. Hence, cellular and extra-cellular structures with sizes in the range of 200-2000 um dominate optical measurements of biological tissues. The contribution from very small structures (<200 um) is still detectable, however, as apparent "Rayleigh scattering" which is significant in tissues with collagen. Two measurements are able to discern the size distribution: (1) Confocal reflectance measurements as the depth position (z) of the focus is scanned down into the tissue, ( ) . (2) Wavelength dependence of diffuse light reflectance. Such measurements offer an opportunity to assess and monitor the nanoarchitecture and microarchitecture of skin and other tissues. Detecting the effects of pathology, actinic damage, and pharmaceuticals on the skin are potential applications of these non-invasive optical methods. About Prof. Jacques Steven Jacques has held appointments at the Wellman Center for Photomedicine and Massachusetts General Hospital, Harvard Medical School and University of Texas. He is currently Professor of Dermatology and Biomedical Engineering at Oregon Health & Science University in Portland, Oregon. His work continues on developing novel uses of optical technologies for both therapy and diagnosis. He developed a hand-held polarized light camera to visualize skin cancer margins and guide surgical excision, tested in clinical trials and licensed to a company. Professor Jacques currently works on bridging between the possibilities afforded by optical technologies and the needs of molecular and cellular biology. Phagocytosis is a spatio-temporally defined multi-step process including ligand-receptor binding and signalling to the cell’s acto-myosin cytoskeleton for engulfment. Quantitative imaging is essential in order to understand better how these processes are coordinated and sensitive to the physical properties of the particle (size, shape, elasticity, ligand density). The successful candidate will image receptors, actin and myosin proteins in phagocytic cups using live-cell fluorescence microscopy of transfected RAW macrophage-like cells, as well as scanning-electron microscopy to quantify cup shapes. He/she will conduct phagocytic assays of bacteria and opsonized particles of different shapes coupled with dissection of the underlying cell signalling. Assays may also be conducted in a flow chamber. The successful candidate will join the lab of Prof. Gadi Frankel (g.frankel(a)imperial.ac.uk) in the Department of Cell and Molecular Biology located on South Kensington campus. The group works in molecular pathogenesis, host-pathogen interactions, and experimental systems biology.
